چکیده

Vitamin A and its analogs (retinoids) have shown great prom ise for the chemoprevention of cancer as well as being a possible new class of chemotherapeutic agents. A Phase I and II trial of 13-c/s-retinoic acid in advanced cancers was initiated, and the clinical pharmacology of the drug was studied. All patients received p.o. 13-c/s-retinoic acid starting at 0.5 mg/ kg/day, escalating over 4 weeks to a maximum dose of 8 mg/ kg/day in divided doses. Although there was a linear correla tion of plasma concentration with dose escalation, large interindividual variations in peak plasma concentrations were noted. At the maximum drug dose, the mean peak plasma concentra tion was 4 x 10~6 M. There was little drug accumulation on this schedule, as trough concentrations between p.o. doses often dropped below 1 x 10~6 M. The drug was metabolized extensively to a metabolite, the concentrations of which ex ceeded parent 13-c/s-retinoic acid concentrations with chronic dosing. Retinol concentrations were below the normal range.

افزایش اثرات درمانی سیس پلاتین و 5- فلورواوراسیل بر روی رده‌های سلولی AGS و KYSE-30 با استفاده از تیمار ترکیبی رتینوئیک اسید تمام ترانس

Backgrounds and Objectives: All-trans retinoic acid (ATRA) which is a derivative of vitamin A, exert fundamental effects on regulation of cell growth, differenation and apoptosis. Recently, resistance to cisplatin and 5-fluorouracil developed in gastric adenocarcinoma and squamous cell carcinoma. In this study, we investigated the combination treatment of ATRA with cisplatin and 5-fluorouracil ...

13-Cis-retinoic acid: pharmacology, toxicology, and clinical applications for the prevention and treatment of human cancer.

Retinoids, particularly 13-cis-retinoic acid, have shown great promise against a number of benign, but serious dermatological conditions. In animal models, 13-cis-retinoic acid functions is a potent antipromoter whether a cancer has been initiated by chemical, physical, or viral agents. Additionally, substantial antiproliferative activity of this compound has been demonstrated in vitro in many ...

The effect of bilateral intrahippocampal injection of all–trans retinoic acid on spatial learning in adult male rats.

Introduction: Previous studies have shown that vitamin A and its derivatives such as retinoid and all-trans retinoic acid have a crucial role in memory, learning and synaptic plasticity. The receptors of vitamin A are seen in different parts of the brain such as hippocampus, where vitamin A has an important role in learning. In this study, the effect of intrahippocampal (CA1) injection of al...

Prophylactic effect of all-trans retinoic acid in an amyloid-beta rat model of Alzheimer\'s disease

Introduction: Retinoid signaling has been argued to have favorable effects on Alzheimer's disease (AD). We studied the role of chronic intracerebroventricular (ICV) injection of all-trans retinoic acid (ATRA) on the amyloid-beta (Aβ) model of AD. Methods: Adult male rats weighing 260-330 g were divided into 12 groups of 8 each. Six groups of rats received ATRA (3nM, 30nM, 3μM, 0.3mM, 30...
